Compatibility of TC-RNA Chemistry with Common Modifications

Abstract

Recently, 2’-O-thiomorpholine-4-carbothioate (TC) was introduced as an alternative protection chemistry to TBDMS in RNA synthesis. TC-RNA chemistry has some significant advantages: one-step deprotection process; coupling efficiencies comparable to DNA chemistry; and has proved to be an efficient means of synthesising ribozymes. Since there are many applications that require modified RNA oligonucleotides, we have looked at the compatibility of TC-chemistry with some common modifications such as cholesterol, FAM, and cyanine type dyes.
